Malaria rapid diagnostic tests (RDTs) are kits used for the diagnosis of malaria. The test is based on the principle of antigen-antibody precipitation, wherein malaria is diagnosed on the basis of the presence of the malarial parasites in the human blood sample. The test is a very simple one, that can be easily carried out in remote areas, as well without the need for any specialized infrastructure or apparatus. The malarial parasite present in the human blood sample acts as an antigen for the test. Malaria is caused by the Plasmodium parasite. A malarial antigen detection test is conducted to diagnose if a particular individual has malaria infection or not. It is easy to cure malaria if it is diagnosed at an early stage. The test is quite simple and does not require any special infrastructure or tools. Thus, it is very easy to diagnose individuals with malaria even in rural areas where laboratory and diagnostic facilities are not very well established.

If malaria is not detected at an early stage, you may experience other symptoms at a later stage of infection that will indicate a malaria test to be done.
To carry out a malaria antigen detection test, the blood is drawn from the patient, usually from the tip of a finger. An antigen detection test is done on a nitrocellulose strip that contains some dye-labeled antibodies. Each antibody is specific for a certain antigen and can recognize the specific species of malaria causing parasite. Therefore, specific nitrocellulose strips are used for specific Plasmodium species. The blood drawn from the patient is mixed with a lysing agent to rupture the red blood cells and release the parasite protein into the fluid. A specific antibody labeled dye is bound to the strip in the test line. The control line is bounded with the antibody specific for the labeled antibody or antigen. The blood and buffer mix with each other. If the blood sample contains parasite protein, it will form an antigen-antibody complex that gets accumulated on the test line.
The nitrocellulose strip contains a window wherein you will be able to interpret the results of your diagnostic test. Generally, there are three sections in the nitrocellulose strip where you can see a red line, these sections are termed as C (control), T1 (test 1), and T2 (test 2). You will be considered as positive for malaria if:
If you get a line at C but no line appears on either T1 or T2, then, in that case, you are not affected by the malaria parasite. Also, if the line appears on either T1 or T2 or both but no line appears on C, then the test is known to be invalid. In such cases, you should repeat the test on a new nitrocellulose strip.
